Abstract:

          Cymbidium aloifolium (L.) Sw. is an epiphytic orchid that has been used as ornamental and medicinal plants. This study aimed to identify the suitable conditions for in vitro seed germination and plant multiplication of C. aloifolium (L.) Sw.. Immature seeds obtained from 10 month scapsules were cultured on ND (New Dogashima, 1993), MS (Murashige & Skoog, 1962), 1/2MS, and 1/4MS medium. ND medium was found to be the best medium for invitro seed germination. Protocorms of C. aloifolium were cultured on liquid and solid ND medium supplemented with various concentrations of BAP and NAA. Analysis of variance revealed that plant multiplication of C. aloifolium was affected by medium states and PGRs supplemented in basal medium. The combination of 0.5 mg/l BAP and 0.1 mg/l NAA supplemented in liquid ND medium provided 100% shoot formation and 6.36 shoots generated from protocorms, while the liquid ND medium added with 0.1 mg/l NAA provided the longest shoot at 6.26 cm. The most effective medium to obtain 98.0% PLB formation and 19.92 PLBs per protocorm was liquid ND medium fortified with 0.5mg/l NAA after culture for 2 months.
